Acanthamoeba-Campylobacter coculture as a novel method for enrichment of Campylobacter species.
In this study, we present a novel method to isolate and enrich low concentrations of Campylobacter pathogens. This method, Acanthamoeba-Campylobacter coculture (ACC), is based on the intracellular survival and multiplication of Campylobacter species in the free-living protozoan Acanthamoeba polyphaga. Four of the Campylobacter species relevant to humans and livestock, Campylobacter jejuni, C. c...
متن کاملProtozoan Acanthamoeba polyphaga as a potential reservoir for Campylobacter jejuni.
We showed by a laboratory experiment that four different Campylobacter jejuni strains are able to infect the protozoan Acanthamoeba polyphaga. C. jejuni cells survived for longer periods when cocultured with amoebae than when grown in culture alone. The infecting C. jejuni cells aggregated in amoebic vacuoles, in which they were seen to be actively moving. Furthermore, a resuscitation of bacter...
متن کاملMultilocus sequence typing and FlaA sequencing reveal the genetic stability of Campylobacter jejuni enrichment during coculture with Acanthamoeba polyphaga.
Low concentrations of Campylobacter jejuni cells in environmental samples make them difficult to study with conventional culture methods. Here, we show that enrichment by amoeba cocultures works well with low-concentration samples and that this method can be combined with molecular techniques without loss of genetic specificity.
متن کاملEnrichment medium for isolation of Campylobacter jejuni-Campylobacter coli.
A broth enrichment medium for the improvement of isolation of Campylobacter jejuni-Campylobacter coli from stool samples and other specimens is presented. Of 1,228 samples examined in parallel, positive results were obtained from 81 by direct inoculation of selective media and from 112 after enrichment. Thus, an increase of 27.7% in the isolation rate was obtained by using the enrichment medium...
متن کاملPrevalence of Campylobacter jejuni and Campylobacter coli in captive wildlife species of India
Campylobacteriosis is an important zoonotic disease and the prevalence of Campylobacter is largely unknown in the wildlife of India. A total of 370 samples, comprising of 314 fresh faecal samples from apparently healthy captive wild animals and birds, 30 stool swabs from animal care takers and 26 samples of the animals’ food and water were collected from G. B. Pant High Altitude Zoo, Nainital, ...
